When you're specifying the part of the day of a specific time, as in "is it at 4 in the morning or at 4 in the afternoon?" we always use the preposition "de". It'd be incorrect or very odd to say for example "El vuelo sale a las cuatro en/por/a la mañana/tarde." (The flight is at 4 AM/PM)
