Hola Emanuel,

the preposition is needed here, not optional. It wouldn't be right to say:

No he salido todo el día.

Saying "en todo el día" has the nuance of "not even once" in the whole day. We use it with sentences normally in negative and with length of time:

- No he visto a Juan en todo el verano.

-Mi jefe no me ha dado vacaciones en todo el año.

-No he puesto la televisión en todo el día. Hoy prefiero tener un día de lectura.
