Dije ________ a la propuesta del jefe.
‘Dije ________ a la propuesta del jefe.‘ I said ‘que sí’ and was marked wrong - isn’t that acceptable? I actually thought ‘que’ would be essential here?

Yes, you're right! - I just added the answer including "que..." to the correct answers.
Yes, it's quite natural to use "dije que sí, dije que no". It's not essential though. If you include "que" it'd be a more "relaxed, colloquial" phrase, and if you say "dije sí" is a bit more literal, but still OK.
